PEGS follows approximately 20,000 adults to discover how the environment and genes affect health. In 2002, PEGS was initially established as the Environmental Polymorphisms Registry (EPR), and the Environmental Polymorphisms Pilot Study (EPPS) was established to assess willingness to participate in a longitudinal cohort. Based on the favorable response rate, enrolment for PEGS began in 2005 by recruiting individuals living in the greater Research Triangle Park region in North Carolina, United States of America, who reflect the overall demographics of the area in terms of age, gender, race and ethnicity. Participants are primarily female (62%) and white (63%), with a mean age of 50 years. All participants are followed up periodically for up to 25 years after enrolment, and approximately 11,000 are actively engaged with annual contact.
